Lecture #1 – Legacy of the Past

A historical perspective covering the emergence of the modern pharmaceutical industry from the ancients to the modern era. The discovery of early medicines from plant sources,the creation of early pharmacopeias with drugs characterized via observations of pharmacology in animals and patients without any knowledge of compound structure or the biological target. The 19th and 20th century, from the dyestuffs industry, bacterial cell culture and the conquest of infectious diseases, the role of pharmaceutical companies during World War 2 and the formation of fully integrated drug discovery entities. Conclude with the impact of two pioneers of medicinal chemistry, James Black and Paul Janssen who were instrumental in the concept of logical drug design.

Lecture #2 – Modern Drug Discovery: Target Selection through Lead Identification

Overview of the small molecule drug discovery process and the phases where medicinal chemistry is key. This will cover phenotypic screening versus target-based screening and be devoted to a single program from initial target characterization of a G-protein coupled receptor, the identification of chemistry starting points, primary and secondary biological assays, the importance of quality lead structures, and their suitability for advancement. This will conclude with a working SAR model that will be used to advance to lead optimization.
